Is wheat gluten peptide gluten-free?

Short answer: No - wheat gluten peptide cannot be assumed to be gluten-free simply because it has been hydrolysed. Gluten-free status is a threshold claim set by regulation, typically expressed in the low tens of parts per million of gluten, and it must be demonstrated by testing the actual ingredient with a method suited to hydrolysed protein. Standard sandwich ELISA kits such as the R5 sandwich format can underestimate gluten in a hydrolysate, because the antibodies need intact binding sites that hydrolysis has already cut. Where a gluten-free claim is required, the material should be tested with an appropriate competitive or otherwise validated method, supported by supplier documentation and batch results.
Why hydrolysis changes the analytical problem
Wheat gluten is the protein fraction that triggers coeliac disease. Hydrolysing it into peptides makes the material more soluble and easier to use, and it can disrupt some of the sequences that antibodies recognize. The difficulty is that the same cleavage also breaks the structures the most widely used test kits rely on. A sandwich ELISA needs two antibodies to bind the same antigen at the same time, which works well for intact protein and poorly for fragments only a few amino acids long. The measurement can therefore read low while gluten-derived peptide is still present.
What "gluten-free" actually means
- United States. A gluten-free claim is defined by regulation and applies to foods that do not contain wheat, rye, barley or their crossbreeds, or that do but have been processed to remove gluten and stay below the defined threshold.
- European Union and UK. A gluten-free claim requires no more than the regulated gluten threshold, conventionally described as 20 mg/kg (ppm), with a higher band for the restricted very low gluten claim.
- Other markets. Canada, Australia and New Zealand and many others set their own thresholds and their own rules about which products may carry the claim.
- Practical consequence. The claim is attached to the finished product as sold, not to the ingredient as supplied. A compliant ingredient does not guarantee a compliant finished product if other components or the line add gluten.
Testing choices for hydrolysed protein
Method choice matters more here than in most protein applications. A sandwich ELISA needs two intact binding sites and can under-report gluten in extensively hydrolysed material. A competitive ELISA is designed for small fragments and is generally the more suitable format, though it needs its own validation. DNA-based methods detect wheat DNA rather than the protein; mass spectrometry can target marker peptides but needs method development and standards. Because the kit changes the number you get, a gluten-free conclusion should state the method, the lot, the limit of detection and the reference material used.
Points that commonly go wrong
- Carrying an R5 sandwich result from an intact wheat protein over to a peptide grade without re-validating the method.
- Assuming a low degree of hydrolysis is "safer" for the claim. Less hydrolysis leaves more intact gluten, which usually means more risk, not less.
- Overlooking carriers, flavours, shared drying and packaging lines that may add gluten.
- Treating wheat starch or wheat fibre as equivalent to a wheat protein peptide; they carry different gluten loads.
How to handle a gluten-free requirement
The most defensible route is usually to avoid wheat as a source entirely: pea, rice and corn peptides carry no wheat gluten, so a gluten-free position becomes a matter of controlling cross-contact. Where a wheat peptide is chosen for a functional reason, plan for a validated method, a sampling plan that covers lots over time, and a claim review against the regulation of each market.
